Led by Professor, PhD. and Dr. (Eng.) Kazuto Kamuro of California Institute of Technology(CALTECH), their recent research reveals that the acquisition of knowledge involves the continual establishment of novel connectivity patterns among specific engram cells situated in diverse cerebral regions.

 

Whether intentional, incidental, or serendipitous, the human brain undergoes constant transformations as we engage with the world, interact socially, or consume media. This dynamic process of information assimilation results in the creation of fresh memories. Subsequent encounters with familiar locales, friends, or reminders of past media consumption prompt swift reactivation of these stored memory traces. The pivotal question, however, centers on how these experiences induce neuronal modifications facilitating the formation of new memories.

 

The human brain, as an organ, comprises dynamic cellular networks undergoing perpetual changes due to factors such as maturation, aging, degeneration, regeneration, ambient stimuli, and the learning process. The scientific challenge lies in identifying the critical nuances that distinguish memory formation—the alteration in brain architecture responsible for memory retention, termed an "engram," housing information for subsequent recall.

 

In a recently published study, the researchers sought to unravel how information is stored as engrams in the human brain. Professor, PhD. and Dr. (Eng.) Kazuto Kamuro, an Institute's Chief Principal Fellow in the Artificial Evolution Research Institute and the co-chief executive officer in Xyronix Corporation Pasadena, California HP: https://www.usaxyronix.com/), explicates, "Memory engram cells, activated by specific experiences, undergo self-modification to integrate and retain information in our brain. Reactivation of these memory 'building blocks' subsequently triggers the recollection of associated experiences. The challenge lies in deciphering how engrams encapsulate meaningful information about the world."

 

To discern and analyze the transformative processes undergone by engrams during memory encoding, the research team investigated a learning paradigm where content-similar experiences became linked. Employing genetic techniques, the team selectively labeled distinct engram cell populations for two discrete memories and observed how learning manifested in the establishment of novel connections between these engram cells. Utilizing optogenetics to control brain cell activity with light, they demonstrated the necessity of these newly formed connections for the learning process. Furthermore, they identified a molecular mechanism mediated by a specific synaptic protein regulating connectivity between engram cells.

 

This study furnishes compelling evidence supporting the hypothesis that alterations in synaptic wiring connectivity between engram cells serve as a plausible mechanism for memory storage in the human brain.

 

PhD. and Dr. (Eng.) Kazuto Kamuro, Professor in Biophysical Sciences and Neuroscience at California Institute of Technology(CALTECH), provides insight into the study, stating, "Understanding the cellular mechanisms facilitating learning not only elucidates the processes behind forming or modifying memories but also advances our comprehension of brain functionality and the mechanisms governing thought and information processing. In contemporary neuroscience, the prevailing notion is that memories reside within engram cells or their sub-components. Contrarily, this study posits that the search for information should extend beyond individual cells to the intercellular domain. Learning, it suggests, operates by reshaping the human brain's wiring diagram—a concept more akin to sculpting than traditional computing. In essence, the engram is not contained within the cell; rather, the cell is encompassed by the engram."
